Notch Signaling

高度保守的Notch信号通路是独一无二的,因为Notch受体及其大多数相应配体(公认的DSL或Delta/Serrate/LAG-2 DSL家族成员)都是附着在细胞表面的跨膜蛋白。因此,Notch信号对相邻细胞之间的相互作用是有限的。

相邻细胞之间的通信至关重要,特别是在细胞的命运和功能尚未确定的早期发展阶段。Notch信号为细胞提供了一种指定自己身份的方法,同时通过侧抑制作用影响相邻细胞的角色和身份。 Notch信号通路的核心包括两个相邻细胞,一个表达DSL家族配体,另一个表达Notch(受体)。当受体与配体相互作用时,两个独立的蛋白酶把North分成为胞外和胞质两部分。ADAM10裂解与其各自配体结合的North余体胞外部分,并被表达细胞的配体吞噬。γ-分泌酶裂解Notch胞质部分。胞质区迁移到细胞核,并在这里与转录因子CSL结合,将其从一个转录阻遏物转化成一个激活物,并上调Notch靶基因的表达。
