Tumor necrosis factor (TNF-)-related apoptosis-inducing ligand (TRAIL, APO-2L) is thought to regulate cell death events in the immune system through TRAIL-R1 (DR4) and TRAIL-R2 (DR5), members of the TNF receptor superfamily. Like the death receptors, including Fas and TNF-R, TRAIL-R1 and TRAIL-R2 have death domains in their cytoplasmic tails that recruit proteins to initiate a cell death signaling cascade. TRAIL (APO-2L) is a 34 kDa type II transmembrane protein. TRAIL forms trimers and is a member of the TNF ligand family. Interaction between TRAIL and TRAIL-R1 and/or TRAIL-R2 rapidly induces apoptosis of TRAIL-sensitive cells. TRAIL can be injected into mice without toxic side effects. Many tumor cells are killed by TRAIL and TRAIL is currently being considered as on of the most promising potential new cancer therapeutics.
