This antibody recognises SIGN-R1, a C-type lectin that has been identified as a murine homologue of the human DC-SIGN related molecule, L-SIGN. Mouse SIGN-R1 is specifically expressed on marginal zone macrophages in the spleen, medullary and subcapsular sinus macrophages in the lymph nodes, and at lower levels on liver sinusoidal endothelial cells. SIGN-R1 has not been detected on murine dendritic cell populations. Like many C-type lectins, SIGN-R1 functions as a pathogen recognition receptor in vivo and plays an important role in defence against pathogens. Antibodies, produced by clone ER-TR9, are reported to block the uptake of neutral polysaccharides by macrophages.

SIGN R1 is a specific marker for the identification of macrophage subpopulations present in the marginal zone of spleen (the so-called marginal zone macrophages (MZM)), in the lymph node medulla, and in some strains, in the peritoneal cavity. MZM of the spleen are involved in the clearance of polysaccharides. Mouse SIGN R1 is a C type lectin, like DC SIGN which is expressed on Human dendritic cells (DCs). However, Mmouse SIGN R1 itself is not expressed on DCs. SIGN R1 exists in an aggregated form, resistant to dissociation into monomers upon boiling in SDS under reducing conditions.
